A mutation which occurs in the promoter region can alter the rate of protein production. This can cause unregulated cell growth and amp up the progress of cancer (Cancer.gov). For example, the 60-year old with cancer might have originally had the same promoters as the non-cancerous 60-year old, but may have suffered from a wide variety of mutations in non-coding regions such as in his promoters causing the "…production of important checkpoint proteins to malfunction. Collectively, these mutations conspire to change a genome from normal to cancerous" (Cancer.gov).
