Ecstasy (MDMA) is a popular drug that produces significant adverse effects, including neurotoxicity. Accordingly, loss of function cytochrome P450 (CYP2D6) alleles contribute to elevated serum levels of MDMA. Locally, MDMA probably increases the release of serotonin, norepinephrine, and dopamine, the sustained exposure of nerve endings to these catecholamines, and in the presence of the met/met variant of catechol-O-methyltransferase (COMT) could contribute to the accumulation of catechol reactive oxygen species. Whether this represents the mechanism for MDMA-induced neurotoxicity will have to await the findings of future research studies.
